      Anyone knows if the school groups those who get into GEP round 2 but not selected for GEP programme in 1 class in P4?

      No. But many will probably end up in the BICEP class.
      Those who do well in either English or Math or both but did not qualify for the GEP programme, the Gifted Education Branch (GEB) will provide the a list to recommend them for high ability programme/s for Math or/and English.

      for Mainstream students, do the HIgh-ability (Maths and English) classes, start in P4 (January) or in P5 ?

                  Only the overall academic for the 1st cut. The strength in languages (esp. Chinese) will come into play during the interview and written test.
                  The overall academic is important as the BICEP class will be the selected mainstream class that will interact with the 2 GEP classes for the next 3 years. They will join the GEP students in classes such as Chinese and PE. The BICEP is predominantly girls while the GEP predominantly boys. PSLE results for BICEP is very competitive with those of GEP as the former is good at Chinese while the latter at Science.
